Description

For quantitative detection of rat WIF1 in cell culture supernates, cell lysates, tissue homogenates, serum and plasma (heparin, EDTA).
Background: Wnt inhibitory factor 1 is a protein that in humans is encoded by the WIF1 gene. The protein encoded by this gene functions to inhibit WNT proteins, which are extracellular signaling molecules that play a role in embryonic development. Additionally, this protein contains a WNT inhibitory factor (WIF) domain and five epidermal growth factor (EGF)-like domains, and is thought to be involved in mesoderm segmentation. This gene functions as a tumor suppressor gene, and has been found to be epigenetically silenced in various cancers.
Principle: Rat WIF1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from Mouse specific for WIF1 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: G29-W379) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for WIF1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substRate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution.
